Spycatcher-Spytag-GFP complex

Abstract

SAXS of 1) Spycatcher003, 2) Spytag003-sfGFP, and 3) Spycatcher003 and Spytag003-sfGFP in complex

Experimental description

The X-ray wavelength was set at λ=1.127 Å and the sample to detector distance was 2100 mm, resulting in scattering vectors, q, ranging from 0.01 Å-1 to 0.47 Å-1. 60 μL of sample with a concentration ~3 mg/ml was prepared in the SEC running buffer. The Shodex KW803 column was equilibrated with a running buffer with a flow rate of 0.65 mL/min. Each sample was injected in an SEC column, and two-second X-ray exposures were recorded continuously for 24 min. BioXTAS RAW was used for further SEC-SAXS processing, including buffer subtractions and merging SAXS frames across the elution peak.
